Hantavirus , which belonging to the genus Hantavirus of the family Bunyaviridae, is an eveloped negative-strand RNA virus with a tripartite segmented genome. The large (L) segment of 6530-6550bp encodes the viral RNA polymerase, the medium (M) segment of 3613-3707bp encodes a glycoprotein precursor (GPC) that is cotranslationally cleaved to yield two surface glycoproteins(G1,G2),and the small(S) segment of 1696-2083bp encodes nucleocapsid protein(NP).The natural host of hantavirus is mainly rodent, and each strain is normally associated with a special rodent host (excluding TPM strain). In generally, the virus causes life-long persistent and asymptomatic infections in rodents, and coevolves with his host. Transmissing of Hantavirus to human is generally via respiratory secretions, saliva and urine and the aerosol route is believed to be more important. Hanta— viruses are known to cause two highly fatal human diseases: hemorrhagic fever with renal syndrome (HFRS) and hantavirus pulmonary syndrome (HPS).There are currently 23 recognised sero/ genotypes , and only I(Ha-ntaan, HTN) and II (Soul, SEO) hantavirus type has been identified in China.Recently, it is reported that PLfU virus has been found in China. The study include two parts:1 -, Lung samples of Rattus norvegicus were analysed by direct immunofluorescence , and 55 positive samples were identified (9.3%). The results of RT-PCR of 4 positive samples (JilinlO5(K JilinlO55> JilinlO56 and Jilinl060) showed that they were II (SEO) hantavirus type. And then a strain , designated as CJilinl050/Rn/2002 , was isolated by Vero-E6 cell.2^ Lung samples of Apodemus peninsulae captured from different epidemic areas of Til in were analysed by directed immunofluorescence and RT-PCR, and 4 postive samples were identified. Two virus strains (CJilin89/Ap/2002 and CJilin93/Ap/2002) were isolated by Vero~E6 cell. Subsequently, Medium (M) genome of CJilin93/Ap/2002 strain (abbreviated as CJilin93) was cloned , sequenced and analysed . The complete nucleotide sequence of the M genome of CJilin93 strain included 3577 bp in length , which encoded a GPC (glycoprotein precursor) of 1135 aa . Sequence comparison revealed that close relation between CJilin93 strain and Baol4 strain carried by Apodemus agrarius. In contrast, the AMR strain harboured by Apodemus peninsulae showed a higher level of genetic diversity from CJilin93 strain based on G2 segment alignment. Phylogenetic analysis also showed that the CJilin93 strain and Baol4 strain formed a common clade. It is suggested that Apodemus peninsulae carried HTN virus in the northeast of China.
