Objective: To investigate the derivants of bergenin through structural modification and their activity of anti-tumor cells. Methods: Given dry pyridine as the solvent and catalyst, acetylation reactions were undertaken with bergenin and different proportions of acetic anhydride; Given dry DMF as solvent and NaH as catalyst, methylation reactions were undertaken with bergenin and CH3I; In vitro anti-tumor activity of bergenin and its six derivants was measured in human tumor cells line using the upgraded MTT means, cisplatin being the positive control. Results: We got 11 derivants of bergenin through structural modification. Bergenin and its six derivants manifested anti-carcinoma activity neither in human leukemic cells (IC5o>100μg/ml) nor in human hepatoma carcinoma cells. Conclusions: Eight new derivants of bergenin that had not been reported were obtained and they manifested no anti-carcinoma activity in human leukemic cells and human hepatoma carcinoma cells.
