Objective: To investigate the role of the titin gene and its influence onthe contraction protein gene such as β-MHC and cTnI in the procession ofBMMSCs differentiating into cardiomyocytes in vitro . Methods: BMMSCs were isolated from bilateral thighbones andtibias of Wistar rats, purified by adhesive-screening method, and expandedin culture for more than 4 passages .They were exposed to 5-aza(10μmol/L)for 24 hours.we observed the morphology of BMMSCs withphase-contrast microscope. Then we transfected the recombination plasmidvector into them.After interference 24 hours,we extract RNA withTripure,Semi-quantitative Reverse transcription-polymerase chainreaction(RT-PCR) detected the expression of titin,β-MHC and cTnI at firstday,forth day,seventh day,fourteenth day after our treatment respectivelyin vitro. Results: 1. we obtained the mild expression of titin,β-MHC and cTnI genebefore 5-aza treament,but the expression of them increased after induction. 2. After recombination plasmid were transfected into BMMSCs whichhave been induced by 5-aza,the expression of titin gene was impaired. 3. The expression of β-MHC gene kept invariant,while the expressionof cTnI gene was weakened after the interference. Conclusion: 1. BMMSCs were a kind of cardiomyogenesis cell. 2. titin gene probably play an important role during the procession ofBMMSCs differentiating into cardiomyocytes in vitro,and it may regulatethe expression of cTnI.
