The Study Of Effect Of NOSI On Childhood's Learning And Memory And Syp, GFAP After Intrauterine Distress

Objective: TO investigate whether intrauterine hypoxia and ischemia can produce long-time effects and whether NOSI can prevent these damges. Methods: Pregnancy rats were divided randomly to three groups.Normal group: 2 rats with gestational age of 17 days were decided to be performed cesarean section on pregnant 19 days.hypoxia and ischemia reperfusion group:2 rats with gestational age of 17 days served as I/R group whose uterine blood supplying were reperfused after clamped for 15 mins.NOSI group:injection of L-NNA 4mg/kg into pregnant rats'abdomen before ischemia following by injection of aminoguanidine(AG) 500mg/kg before operation.Cesarean section was performed on pregnant 19 days.Thereafter pups were given to surrogate mothers and examined the ablity of learning and memory at 40 day of age after delivery.Then the water maze test was performed to detect the space learning ability and memory function of rats;and the changing of synaptophysin levels in hippocampus were detected by immunohistochemical staining. Result: Behavioral results show that fetal distress produces cognitive impairment demonstrated by Morris water maze performance including (i) a higher escape latency score and (ii) a decreased cross platform time.The COD of Syp and GFAP positive immunoreactive product in hippocampus were less decrease than the normal group or NOSI group. But the behavioral results and the COD of Syp and GFAP were no difference between normal group and NOSI group. Conclusions: Fetal distress produces cognitive impairment and leads to the decreasing of Syp and GFAP in hippocampus.Effective measure can relieve these damages.
