| Food is very important with the mankind surviving and developing. Thefood safety is concerned by the society with the developing in food industry.The food poisoning is the main problem in which the food poisoningbacteria is dominating. Some toxin including SE ,Vero toxin,BoNTs cancause serious food poisoning. With the developing of the molecule biotech,the study on food poisoning toxin is increasing and the cognition isenhancing. In the diagnosing and detecting ways, the toxin gene is morecredibility and sensitivity sign than others. The detection ofpoly-recombined toxins may be an innovation which will be morebroad-spectrum and quicker.In this experiment we studied S.aureus ,coli.O157:H7 and clostridiumbotulinum. We designed and synthesized the primer according the genesequences which were published in the articles or Genbank. Amplifying thegene fragments which include SEA,SEB,VT1B,VT2B and C-fragmenton BoNTa。After purified, the gene fragments were ligated with pUCm-Tvector and transformed into E.coli DH5a. The homology was 100% bysequencing.The cloned gene fragments were ligated in series with the linkersequences. We abstained an sole-peptide-chain, in which there were tworestriction enzyme sites NcoI,XhoI. The primers was designed andsynthesized that the ending codon was TAA. The full length sequence wasamplified through Overlap Extension PCR, then ligated with pET-28a andtransformed into E.coli DE3. The homology was 99% that the gene... |
