Objective:The thesis is to explore the protective effects and mechinisms of octreotide on the multiple organ injury caused by acute necrotizing pancreatitis (ANP) of rats. By observing changes of expression of aquaporin-1 (AQP-1) on ALI caused by ANP of rats, the thesis is also to explore the relationship between aquaporin-1 (AQP-1) and acute lung injury (ALI) and to investigate the possible effects and mechinisms of octreotide. Methods:Twenty-four wistar rats were randomly divided into three groups (n=8): sham operation group (SO group), ANP group, and octreotide treatment group (OT group). The ANP model of rats was established by retrograde injecting 5% sodium taurocholate solution through biliary-pancreatic duct. The OT model of rat was established by subcutaneous injecting octreotide (10μg/kg). The changes of pancreatic, hepatic, pulmonary, small intestine, and renal morphological damage were observed with light microscope in each group. Try to measure the changes of lung wet/dry (W/D) and ascites/avoirdupois weight ratio; Try to measure the spleen T lymphocyte transformation rate (LTR) by colorimetry; Try to measure the changes of numbers of red blood cell (RBC), the hemoglobin (Hb) concentration and hematocrit (HCT); Try to measure the changes of the serum amylase (AMY),alanine transaminase (ALT),blood albumin (ALB),alkaline phosphatase (ALP),blood urea nitrogen (BUN), the contents of serum creatinine (CRE), blood sugar (GLU),serum calcium (Ca2+); Try to measure the changes of tumor necrosis factor (TNF-α) and interleukin-6 (IL-6) concentration; Try to analyze the changes of nuclear factor-kappa B (NF-κB) expression in the pancreas, liver, kidney and small intestine of rats in each group by immunohistochemistry. Try to analyze the changes of AQPl expression in the lungs of rats in each group by immunohistochemistry, reverse transcription-polymerase chain reaction (RT-PCR) and western blot technique.
