Object: To develop a new ex vivo rat model of lung perfusion for the lung ischemia research and study the effect of Vitamin C on the lung is-chemic-reperfusion injury using this new model.Method: 32 pairs of SD rat were divided into 4 groups: 8 pairs in each group; one of each pair was used as donor to provide lungs, and the other rat was used for perfusion as recipient. The lungs were connected with the perfusion circuit after 6 hours preservation at 4°C and perfused for 1 hour. The pulmonary artery pressure (PAP) and peak inspiratory pressure(PIP) were dynamically measured. And the wet/dry ratio, MDA, ATP, PaO2 and histopathologic alternation were analyzedResult:This new model worked well. The operation time of the donor and recipient rats were 30 and 25 minutes. Compared with group A, the group B C D had the the lower water content MDA PAP (P<0. 05) , the higher ATP CP<0. 05) , and the less histologic injury after reperfusion. PIP in group A B C were almost the same, but PIP in group D was lower than the other three (P<0. 05) .Conclusion: This new ex vivo rat model of lung perfusion for the lung ischemia research has many advantages: less cost, simple technique, stable result. VitC has protection effect on the lung ischemic-reperfusion injury.It can reduce the pulmonary vascular resistance and the production of oxygen free radicals, improve the level of ATP and the oxygenation ability, and alleviate the pulmonary edema and histological injury. Increasing the concentration of VitC in the preservation solution maybe provide better protection to the lung.
