Objective: To explore the combined effects of arsenic trioxide and all-trans retinoicacid on the inducement differentiation, proliferation and apoptosis of NB4, R4 cell linesof acute promyelocytic leukemia, and to provide a theoretical basis for the combinationof arsenic trioxide and all-trans retinoic acid in the treatment of APL.Methods: To investigate the combined effects of arsenic trioxide and all-trans retinoicacid on the inducement differentiation, proliferation and apoptosis of NB4, R4 cell linesof acute promyelocytic leukemia, the differentiation was examined with NBT, CD11b,proliferation by MTT, apoptosis by flow cytometry, mRNA of PML/RARa withRT-PCR, the fusion protein of PML/RARa by Western blotting.Results: 0.5 μ mol/L As2O3 synergized with 0.1 μ mol/L ATRA to induce apoptosisand inhibit proliferation, and decreased the differentiation in NB4. however, 0.5 μ mol/LAs2O3 synergized with 0.1 μ mol/L ATRA in the inducing apoptosis, differentiation andinhibiting proliferation on R4.Conclusion: The study indicated that 0.5μmol/L As2O3 synergized with 0.1μmol/LATRA to induce apoptosis and inhibit proliferation, however, 0.5μmol/L AS2O3synergized with O.lumol/L ATRA in the inducing apoptosis, differentiation andinhibiting proliferation on R4. We draw the conclusion that the better effect may bemade by the combination of arsenic trioxide with all-trans retinoic acid in the treatmentof APL.
