Objective—To study the effects of NSCs different implantation site on the behavioral recovery and secretion of neural growth factor after MCAO damage in rats and select the best implantation site.Methods—To isolate , culture and passage NSCs from embryonic rat brain, then transplant them into MCAO rat brains.The rats were divided into 6 groups: about 5 μ l of cell suspension(8 × 10~5 NSCs) prelabeled with Brdu were transplantated into the hemisphere 1 week after MCAO respectively (experiment group Ⅰ : the ipsilateral striatum , Ⅱ: the ipsilateral cortex,Ⅲ: the ipsilateral ventricular,Ⅳ: the contralateral striatum); sham grafted rats received 5 μ1 of normal sodium chloride 1 week after MCAO ; Sham-operated rats received 5 μ of normal sodium chloride. All rats were compared in behavioral test 24 hour, 1 month , 2 month and 3 month after transplantation respectively. Ultrapathologic examination was performed 2 months after transplantation. The pathologic examination also was performed and the secretion of neural growth factor was measured 3 months after transplantation.Results—The cultured NSCs showed the expression of Nestin antigen and were able to differentiate into neurons and astrocytes in vitro.There are no significant differences between the experiment groups and the sham-grafted group 24 hour and 1 months after transplantation. Rats in experiment group I showed significant improvement on behavioral test compared with other experiment groups and the sham-grafted group 2 months after transplantation. Rats in experiment group Ⅰ and Ⅲ showed significant improvement on behavioral test compared with the other experiment groups and the sham-grafted group 3 months after transplantation.The pathology proved that there were Brdu positive cells in the slides of transplanted groups, and the cells could differentiate into neurons and...
