As an important indicator of heart-related disease, triglycerides (TG) have already aroused people's attention with the raise of living standard in recent years. Different techniques for the determination of TG have been developed and applied in clinic laboratories.The quantitative analysis of TG is a little difficult because the composition of TG in serum is complex. A standard and reference method is needed.In this thesis, an IDGC/MS method was developed to determine total glycerides, pure triglycerides and free glycerol in serum. [13C3] glycerol and [ 13C3]tripalmitin synthesized from[ 13C3]glycerol were selected as labeled materials in the research. The conditions in serum sample pretreatment, extraction of TG, derivation by heptafluorobutyric anhydride, and the operation parameters of GC/MS were studied.At the optimized condition, the RSD of the total glycerides and free glycerol of four human serum samples and the pure triglycerides by the ID/MS are less than 2%.
