Objective:To investigate the effect of the ligand of PeroxisomeProliferator-actived Recepter gammar Rosiglitazone using alone or combinated with Cisplatin on human lung adenocarcinoma cell in vitro, and its mechanism.Methods:human lung adenocarcinoma A549 cell line was cultured invitro, A549 cell line was treated with various concentration of RSG or CDDP or both, Cell counting method was used to test the influence of growth curve in A549 cells, cell proliforation was evaluated by MTT assay ,The Apoptosis of A549 cells was determined by AO/EB double fluorescence stain and PI staining flow cytometry and cell cycle was analyzed using flow cytometry. The expression of PPAR Y , NF- k B, Bcl-2, Bax protion were investigated by immunohistochemical techniques .Results: (1) MTT assay indicated that RSG whose concentrationexcessed 50μmol/L could inhibit the proliferation of A549 cells in a concentration dependent and time dependent positive;RSG at concentrations 1. 25, 2.5, 5μmol/Lwas not of cytotoxicity to A549 cells (IR<10% ) , but its significantly increased the cytotoxicity of CDDP to A549 cells;these results is consistent with the results of Cell counting method drawing growth curve in A549 cells . (2) AO/EB double fluorescence stain and PI staining flow cytometry data showed the apoptosis of A549 cells induced and cell cycle was arrested at G1 stage by RSG in a concentration dependent manner;RSG(1.25μmol/L) significantly promoted CDDP(1.96 , 2.8 , 4.0mg/L)to induce A549 cells apoptosis.
