Objective To investigate the inhibitory growth effect of recombinantapoptin eukaryotic expreesion plasmid on human ovarian cancer xenografts in nude mouse modle.Methods The recombinant eukaryotic expression plasmid pcDNA-VP3 wassuccessfully constructed. Approximate 2x10~6 cells of ovarian cancer cells (COCl) were implanted subcutaneously into per nude mouse. Twelve female Balb/c nude mice were divided into as follows four groups: High doesage group and low doseage group were received intratumoral injection at does of 100 μg and 50 μg pcDNA-VP3 plasmid cDNA respectively.Plasmid control group and lipofectamine control group were received injection at does of 100 μg pcDNA3.1(+) cDNA and the same volume of liprofectamine.The length and wideth of the xenograft tumor were mearsured every threee days, caculate the volume of the tumor according to the formula:V=LxW2x0.52. These mice were sacrificed after twelve days,and their tumors were excised .The mean tumor weight per group were caculated.RT-PCR was used to detect mRNAexpression of VP3 gene .Apoptosis of the xenograft tumor was observed by TUNLE assay analysis.Resu I ts (1) Recombinant eukaryotic expression plasmid pcDNA-VP3 wasconstructed successfully.(2) The tumor growth inhibitory rate of high dose of pcDNA- VP3 group was76%(p<0.05).The tumor growth inhibitory rate of low dose of...
