Objective: To investigate the proliferation inhibitory effect of Human colorectal cancer cell line SW-480 by Genistein in vitro and to explore its antitumor molecular mechanism.Method: Human colorectal cancer cell line SW-480 was cultured in vitro and used to perform experiments. The antiproliferative effects of Genistein were examined by 3-[4,5-dimethylthiazol-2-yl]5-diphenyl tetrazolium bromid(MTT) assay in vitro.The influences of Genistein on cell growth were measured by counting cell numbers.The influences of Genistein on apoptosis and cell cycle were also detected in SW-480 cells using flow cytometry(FCM).Immunocytochemical staining were used to detect the expression of Bax, Bcl-2, NF-κβ, COX2.Westein bloting was used to analyze the expression of ERK, to elucidate the possible mechanism of apoptosis induced by Genistein in SW480 cells. Result:1. Genistein (of 6.25,12.5,25,50,100μmol/L ) inhibited the growth of SW-480 cells in a concentration-dependent manner.2. Flow cytometry analysis revealed that Genistein arrested cell cycle in G2/M phase and induced apoptosis in SW480 cells.3. Immunocytochemical staining and morphometric quantitative analysis indicated that down-regulation expression of bcl-2, NF-kB, COX2 protein (p < 0.05),while up-regulation in expression of Bax (p < 0.05).4. Simultaneously, change of activation of ERK induced by GEN from 6.25 to 100 jimol/L showed a concentration -dependent manner(p < 0.05).Conclusion:1.
