PURPOSE To investigate the effect and mechanism of the Genisteinderivative 5,4'-Di-n-octoxyl-7-gem-difluoromethylenegenistein(DOdFMG) on the growth and apoptosis of human ovarian cancer(COC1) cells line in vitro.METHODS (1)COC1, COC1/DDP and CHO -K1 cells lines were treatedby Various concentration of DOdFMG for 48 hours.MTT assay was used to determine the effect of DOdFMG on proliferation of COC1, COC1/DDP and CHO -Kl cell. (2) AO/EB fluorescence staining discovered whether DOdFMG could induce apoptosis in COC1 . COC1 /DDP and CHO-Kl cells lines. (3)PI stain flow cytometry (FCM) was used to analyse the apoptosis and cell cycle change of COC1 and COC1 /DDP treated with DOdFMG at 20μm o 1 /L for 24,48,72 hours . (4) FITC indirect fluorescence labeling FCM method was used to analyse proteins expression of NF-kB , Bcl-2 , Bax and Cox2 inCOCl cells treated with DOdFMG;RESULTS Vaious concentration of DOdFMG significantly suppressed proliferation ofCOCl andCOCl/DDP cells lines in a dose-dependant manner. The IC50 was 15μmol/L and 20μmol/L respectively.The IC50 of CHO-Kl was 75μmol/L .The effect of DOdFMG on CHO-Kl cells line was weaker.Analysis using AO/EB fluorescence staining and flow cytometry showed that DOdFMG induced apoptosis inCOCl cells andCOC1/DDP cells. DOdFMG could not significantly arrest cell cycle .DOdFMG downregulated the expression of NF-kB , Cox2 and Bcl-2 proteins and reduced the ratio of Bcl-2 to Bax .CONCLUSION (1) DOdFMG selectively inhibited the proliferation...
