Objectives:(1) To investigate the methodology to analyze DNA stemline ploidy of hepatocyte and hepatocellular carcinoma in the tissue section.(2) To compare and analyze DNA stemline ploidy and parameters of the normal hepatocyte and the hepatocellular carcinoma in adult.(3) To analyze the clinical pathologic significance of DNA stemline ploidy and its heterogeneity in the hepatocellular carcinoma.Materials and methods:The slices of 15 normal hepatocyte patients and 45 heptocellular carcinoma ones were selected, the slices of 4μm, 10μm of which were prepared respectively. DNA stemline ploidy and its parameters were determined with TIGER 920G image cytometry in the tissue section. The optical density of nuclear DNA in the 4μm slice was measured, whereas the volume of the integrated nuclear in the lOum was measured. The total DNA of integrated nuclear volume was calculated by TIGER 920G image cytometry, and the lymphocyte in the same slice was looked on as the control, the value of its DNA stemline ploidy was calculated.Results:(1) In adult, the major DNA stemline ploidy of the normal hepatocyte was diploid (73%). The values of DNA stemline ploidy, the mean volume of the karyon and the rate of dinucleus in the hepatocellular carcinoma were different statisticallyfrom those of the normal.(2) The DI of hepatocellular carcinoma was different from 0.86 to 9.32. The heterogeneity rate of DNA stemline ploidy of the 45 hepatocellular carcinoma was 77.8%. The rate of DNA stemline aneuploidy was 75.6%, and none was DNA stemline diploidy tumor.(3) There was statistical relation between DNA stemline ploidy and its heterogeneity and the volume, the histology classification, the lymph metastasis, the AFP, and the 5cERa.Conclusions:(1) It is feasible and preponderant to use TIGER 920G image cytometry to detect DNA stemline ploidy.(2) Analysis on DNA stemline ploidy and the morphological parameters of karyon are important indexes to identify the hepatocellular carcinoma from the normal.(3) Analysis on DNA stemline ploidy and its heterogeneity is valuable to the pathologic diagnosis, the estimation of the malignant grade and the therapeutic effect and the judgement of the prognosis of hepatocellular carcinoma.
