Objective: To investigate whether stent coated with CD34 antibody can alleviate in-stent stenosis of artery injured by balloon in experimental rabbits. Methods: 1.Gelatin was chosen as the substrate to coat stent, then the stent coated CD34 antibody and manufactured antibody stent. 2. The left and right femoral arteries of New Zealand white rabbits were injured by balloon, and established the animal model. then stents were released in two arteries, antibody stent was released in the left, common stent in the right. One rabbit was sampled at 14 days, and three rabbits at 45 days, after angiography, the arteries where stent in and its two sides were harvested for pathological analysis, measured the diameter of stent(SD) and the most stenosis of the diameter of stent(MNSD) and calculating the most stenosis percentage (MSP). 3. Parameters: ①MSP=(SD-MNSD)/ SD×100%;②neointima percentage of stented arterial segments; ③the neointima in the proximal and distal arteries of stent; Results: 1.At 14 days, the neointima of artery was more obvious in common stent than in antibody stent; 2. At 45 days, the MSP within stent was obviously larger in common stent than in antibody stent (47.03±11.89 vs 16.13±3.93, P<0.05). Meanwhile, the percentage of neointima in common stent was obviously larger than in antibody stent (66.83±6.00 vs 49.64±6.40,P<0.05)also ; 3. The neointima in the proximal arterial segment outside of stent was similar between the 2 kinds of stents, but the neointima in the distal arterial segment of antibody stent was less as comparison with common stent. Conclusion : 1.Gelatin as substrate to coat stent is better than low molecul Dextrose glycoside to absorb CD34 antibody. 2. The antibody stent can alleviate in-stent stenonsis of artery injured by balloon.
