Objective:To investigate the protection effect of gingko bilobaextract in neonatal rat after hypoxic-ischemic brain damage byobservation the PAF level in brain issue of fetal rat with HIBDusing different dosage EGb injection.Methods: (1)For preparation of HIBD model, on postnatal day 7, thepups were anesthetized by ether inhalation, then the left commoncarotid artery was ligated. After surgical procedure, the pupswere allowed to recover for 2 hours, then exposed to a gas mixtureof 8% oxygen in 92% nitrogen for 2 hours. The Sham control groupsubjected to neither ligation nor hypoxia.(2)EGb treatment: Treatedgroups were set up by different EGb of 25mg/kg/dose,50mg/kg/doseand 100mg/kg/dose.After hypoxia, three kinds of dosageEGb,repectively,were injected intraperitoneally in differenttreated groups.(3) Measurement of brain issue's PAF level andhippocampal area: PAF level was measured by Elisa at 2h,4h,and 6hafter the end of hypoxia-ischemia;To quantify injury severity,hippocampal area was measured using Image software.(4) Statisticalanalysis: ANOVA was used for continuous parameters between groupsin split-plot design. All the data expressed as X ±s. P<0.05 wasconsidered statistically significant.Results: 1. Measurement of PAF: PAF increased in all treated groupscompared with sham control group, but decreased in comparison withHI group. Between treated group and non-treated group, there issignificant difference. In all treated groups, the PAF level in50mg/kg/dose of Ebb was the lowest, which decreased compared withtwo other groups(P<0.01);Between 25mg/kg/dose and 100mg/kg/dosegroups, there is no singificant difference(P>0.05). 2.Hippocampalarea: Area was the largest in sham control group, but the smallestin HI group and there is significant difference between treatedgroups and non-treated groups(P<0.01).Area of 50mg/kg/dose groupincreased obviously in comparision with two other treatedgroups(P<0.01, P< 0.05).But there is no significant differencebetween 25mg/kgand 100mg/kg groups.Conclusion: EGb injected intraperitoneally has protective effectson neurons of hypoxic-ischemic damaged rat, and EGb of50mg/kg/dose makes the best therapy effect.
