OBJECTIVE:To study effects of Panax notogineseng saponins(PNS) on the apoptosis of human acute promyelocytic leukemia(APL) cell line HL-60 and expression of the apoptosis-associated genes p53, bcl-2. METHODS:HL-60 cells were cultured in vitro and treated with 0-1600μg. ml-1 PNS for 4872h. The morphology of the cells was observed by microscopy .The cell growth and proliferation were measured by MTT. The apoptosis peaks were detected by flow cytometry. The apoptosis-associated genes p53, bcl-2 were detected by immunohistochemical technique. The DNA break was examined by Agarose gel electrophoresis.RESULTS:HL-60 cells treated with 800-1600μg. ml-1 PNS for 72h showed the apoptotic morphology changes in microscope. The growth of HL-60 cells treated with 200-1600μg.ml-1 PNS for 48h was obviously inhibited in concentration-dependent manner. 200-1600 μg.ml-1 PNS could induce HL-60 cells apoptosis and the rate of apoptosis increased when concentration of PNS increased. The expression of apoptosis gene p53 significantly up-regulated(p<0.05) and the expression of anti-apoptotic gene bcl-2 significantly down-regulated(p<0. 05) when HL-60 cells were treated with 800, 1600μg.ml-1 PNS for 72h . HL-60 cells treated with 800, 1600μg.ml-1 PNS for 72h showed DNA ladders . CONCLUSION:1. PNS (200-1600μg.ml-1) can induce apoptosis of HL-60 cell2. The apoptosis is related to the up-regulation of apoptotic genes p53 and down-regulation of anti-apoptotic genes.
