The Different Expression Of Metabotropic Glutamate Receptor 1 α(MGluR1α)in The Different Brain Areas Of The Kindled Epilepsia Models Of Rats Induced By Coriaria Lactone (CL) | | Posted on:2005-07-20 | Degree:Master | Type:Thesis | | Country:China | Candidate:X Lin | Full Text:PDF | | GTID:2144360155973230 | Subject:Neurology | | Abstract/Summary: | PDF Full Text Request | | Purpose:1.To evaluate the significance of establishing the kindled epilepsia models of rats induced by Coriaria Lactone(CL)for the epilepsy research2.To study the different expression of Metabotropic Glutamate Receptor 1 a (MGluRl a )in the different brain areas of the kindled epilepsia models of rats induced by Coriaria Lactone(CL)Methods: 30 healthy male SD rats were randomized into 3 groups and received0.75,1.0 and 1 .25ml/kgCoriariaLactone(CL)mtramuscular injection respectively.5 same rats received saline 1.0ml/kg intramuscular injection as the control group.These injections repeated every 84 hours .After injection for 25-30 times,the rats whose seizures reach 3-5grade(Racine scale)within 24 hours after CL injection and have 10 or more seizures between 2 injections are the kindled rats.The rats that do not reach the above conditions arethe non-kindled ones, the EcoG of all groups were recorded with the multi-electrode elecophysiology recorder to view whether the EcoG of kindled rats accord with their seizures and difference among the EcoG of 3 groupsQtindled, non-kindled and control).The kindled rats received CL injection for 30 days after kindled. The brain tissue of all groups were collected and fixed with paraformaldehyde and embeded in paraffin. The coronary Sections of brain tissue were HE and MGLUR1 a immunohistochemistry stained and observed under light microscopy. The positive expression of MGluRl a were "point counted" and the expression numbers were calculated.Results: 1.13 rats were kindled. The kindled rats had all grade(Racine scale)seizures after CL injection and seizures accord with their EcoG Features2. The MGluRl a expression of kindled rats in hippocampus and temporal cortex outside hippocampus was stronger than that of non-kindled and control. (p<0.05).The strong expression was no obviously different between neuron and gliaConclusion: l.The seizures of kindled rats induced by coriaria lactone(CL)accord with their EcoG features. The mechanisms of the kindled model are similar to that of human epilepsy. This is an optimal model for epilepsy research2.The MGluRl a expression of kindled rats in hippocampus and temporal cortex outside hippocampus was stronger than that of non-kindled and control.
