Objectives To examine the differential properties of mucous oligosaccharides in acrolein-induced rat airways and the role of dexamethasone and fudosteine in the regulation of rat airway mucin secretion. Compare the in vivo characteristics of oligosaccharides expression after the administration of dexamethasone and fudosteine.Methods Rat models of mucus hypersecretion of airways were established by exposing to 3.0ppm acrolein fog 6 hours per day for 12 days. Rats were also injected intraperitoneally with dexamethasone (0.5mg.kg~-1.d~-1) or orally administrated with fudosteine (20mg.kg~-1.d~-1, 40mg.kg~-1.d~-1 or 100mg.kg~-1.d~-1) with or without exposing to acrolein fog. The mRNA of MUC5ac and the mucous oligosaccharides in rat airways of each animal were detected by RT-PCR and lectin histochemistry. The results were processed semiquantitatively by image analysis. ELISA was employed to detect the release of TNF- α in BALF .Results Compared with control group , 3.0ppm acrolein significantly increased the concentration of TNF- α and cytologic count in BALF ,and the expression of MUC5ac mRNA, α NeuNAc2,3Gal Sialic acid, α-L-Fuc, α -NeuNAc2,6Gal Sialic acid in rats airway (P<0.05) . Both dexamethasone (0.5mg/Kg) and high dose of fudosteine (100mg/Kg) significantly decreased the levels of TNF- α and MUC5ac mRNA expression((P<0.05) and the IODof a NeuNAc2,3Gal Sialic acid, a -L-Fuc, a -NeuNAc2,6Gal Sialic acid in rats airway; But adjusted with the levels of the expression of MUC5ac mRNA, only the expression of a NeuNAc2,3Gal Sialic acid could be decreased in Fudosteine group while a NeuNAc2,3Gal Sialic acid and a -L-Fuc be decreased in dexamethasone group . There was no a -D-Man expression in all groups.Conclusion In acrolein-induced Rat models of mucus hypersecretion of airways , not only the increase of the quantity of mucin but the change of the component of oligosaccharides was observed. Both dexamethasone and fudosteine decreased MUC5ac mRNA expression, however only the expression of a NeuNAc2,3Gal Sialic acid be decreased in fudosteine group. Increased inflammatory cells infiltration and TNF- a may involve in the regulation of the expression of mucous oligosaccharides .
