[Objective] To detect the expression and distribution of Axin in the normal liver tissue ,adjacent tissue of cancer and primary hepatocellular carcinoma tissue and investigate the relationship between Axin expression and the genesis of HCC,and the prognostic factors of HCC.[Methods] The expression of Axin protein was evaluated in paraffin -embeded tissues by immunohistochemical techniques (SABC) in 40 case of HCC,21case of paracarcinomatous tissue and 14 case of normal liver tissue .And 5 case of normal stomach tissue and 5 case of gastric carcinoma tissue was evaluated as a positive control. [Results] The expression of Axin protein was observed in HCC tissues, paracarcinomatous tissues and normal liver tissues. The positive expression of Axin were 57.5% , 90.5% , 92.9% respectively. The brown granules of positive cell were mainly distribted in the cytoplasm. No significient difference in the expression of Axin was found between normal liver tissues and paracarcinomatous tissue(p>0.05),but not between the paracarcinomatous tissue and HCC(p<0.05). Axin protein expression were not related to age,sex ,tumor size and the expression of AFP, but were significantly correlated with portal vein invasion and clinicalpathological stage.In normal stomach tissue ,the expression of Axin in the stem cells is strong ,and weak in the mature cells.But its expression in normal liver tissue is well-proportioned. [Conclusion] Axin expresses mainly in the stem-cells,and itsoexpression is important to sustain the function of the stem-cell; Hepatitis B and liver cirrhosis have no influence to the expression of Axin;The reduced expression of Axin correlates with the genesis of HCQThe check of its expression can be used as an independently index to evaluate the biological behavior and prognosis of HCC.
