Experimental Study Of The Effect Of Vascular Endothelial Growth Factor On The Cultured Alveolar Epithelial Cells â…¡ In Vitro

Objective It is good known that the vascular endothelial growth factor is angiogenesis,is it only angiogenesis ?if has it other affect Tthese are not quite clear at present .we begin about study the effect of VEGF en development of lung . especially investigate the effect on the alveolar epithelial cells Ⅱ. Whether does VEGF increase surfactant protein (SP) expression and pulmonary surfactant synthesis and secret.Method Wistar rat at 21 days gestation paunch to get embryo, Alveolar Epithelia Cells Ⅱ of the different digest method separate ,purify ,identify and primary culture. Experiment grouping:①VEGF-165 grouping : VEGF-165 6ng/ml,②Dexamethason grouping: Dexamethason 10^-7 mmol/L,③VEGF-165 and Dexamethason grouping: VEGF-165 6ng/ml and Dexamethason 10^-7 mmol/L, ④DMEM contradistinguish. Expression of VEGF,Flk-1, Flt-1,SP-B are measured by SABC of immunology histochemistry. Content of total Phospholipid,lecithin, Sphyingomyelin,lysophosphatidy lglycerin are measured. by thin layer chromatography.Result purity of Primary Culture of AECⅡ reach percent of 89, activity achieved percent of 91.85.Method of different concentration trypsin separate lung tissue , purity ,activity,production of AEC Ⅱ satisfy demand of researching their function in vivo. VEGF-165 grouping total phospholipids (μg/ml):119.8±5.1, 58.7±2.1 29.5±0.58 6.1±0.2,VEGF,Flk-1, Flt-1,SP-B are positive expression; Dexamethason grouping total phospholipids(μg/ml):84.1±10.3,41.1±4.04, 22.6±1.05, 4.97±0.12,VEGF,Flk-1, Flt-1 nagative expression ,SP-B positve expression;VEGF-165 and Dexamethason grouping total phospholipids(μg/ml)143.7±7.75 , 56±2.25 ,32.2±2.01,7.0±0.15,VEGF,Flk-1, Flt-1,SP-B are positive expression; contradistinguish grouping total phospholipids(μg/ml): 43.4±5.1,25.8±1.52,14.3±1.02,3.97±0.15, VEGF ,Flk-1,Flt-1 are positive expression, SP-B is negative expression.Conclusion singleness and united using VEGF-165 and Dexamethason increase synthesis of phospholipids and expression of VEGF, Flk-1,Flt-1, SP-B of AEC Ⅱ are positive; Dexamethason restrain expression of VEGF, Flk-1,Flt-1 ofAECD, hastening synthesis of phospholipids and SP-B expression .it is indicate that VEGF can hasten developing of AEC II, it can inhibit the affect of Dethamethason of down expression of VEGF ^ Flk-1, Flt-1 on AEC II.
