| Objective : To observe the effect of the antithrombin-III on the cytokines and ET in rat with SIRS or/ and MODS. Methods: 70 Wister rats were randomized to three groups : normal control group (n=10 ) , model group (n=30 ) and treatment group (n= 30 ). The model group and the treatment group were given endotoxin two times to make MODS models. The first time each rat was given endotoxin by dose 100ug/kg , 12 hours late 200ug/kg endotoxin was given to each rat by veins . After two hours the treatment group were injected AT-Ⅲ ( 25u/kg ) by veins . Four hours after therapy , all of the rats blood samples were collected . Then the plasma and the serum were separated from the blood samples to assay the levels of IL-1, 6, TNF-a and ET .The lungs , livers and kidneys were collected and reserved in formalin liquid . After dyed by HE, all of the organs were made into pathologic sections to observe the changement under the microposcopy . Result : The concentrations of IL-6, TNF-a and ET (P<0.05)were lower in the treatment group than in the model group and the level of the IL-1 (P < 0. 01 )was significantly lower in the treatment group than in the model group . They had the statistic significance . From the sections we foundpneumonorrhagia^ alveolus atrophy > swellen liver cell and renal cell and many inflammatory cells in the model group . But in treatment group we only found a few inflammatory cells . Conclusion : AT— III could significantly decrease the blood concentrations of IL—1, 6^ TNF-cc and ET so that it reduced the inflammation response induced by LPS. AT-III had the role of inhibiting the forming of thrombus , then it could reduce the damage of lacking of blood and oxygen . AT—III also could control the inflammatory response to decrease the damage of the tissues and the organs through more than one way, so it could inhibit the development of SIRS/MODS .reduce the mortality rate and improve the life quality .
