ObjectiveTo study the expression of bcl - 2 in brain after injection pLXSN - bcl - 2 in rats with focal ischemia - reperfusion via internal carotid artery and its effect on neuronal apoptosis and the expression of caspase - 3 and infarct volume.Methods51 healthy adult Wistar rats were divided randomly into control group , plas-mid control group , and bcl - 2 group, each group including ischemia - reperfusion 9 hours ( n = 6) , and 24 hours ( n = 11) two subgroups. The left middle cerebral artery was occluded for 120minutes, pLXSN and pLXSN - bcl -2 were injected respectively via internal carotid artery 3 hours later. The infarct volume after ischemia - reperfusion 24 hours , expression of bcl - 2 and caspase - 3 , and apoptosis in neural cells were confirmed respectively by staining with 2%2, 3,5 - triphnyltetrazolium chloride ( TTC ), immunohistochemistry, in site cell apoptosis detection.Result1 Infarct volume after ischemia - reperfusion 24 hours of each group was 214.73 32. 15mm~3,221. 56 33.86 mm~3,162. 43 25.46 mm~3. Infarct volume ofbcl -2 group was reduced significantly.2 To detect percentage of positive bcl - 2 cells of each group by immunohis-tochemistry, the result was the percentage of bcl - 2 group , including ischemia-reperfusion 9 hours and 24 hours, was increased significantly.3 The expression of caspase - 3 in each group of ischemia - reperfusion 9 hours was similar. After ischemia — reperfusion 24 hours , the expression of caspase -3 of bcl -2 group was reduced significantly.4 The apoptosis in neural cells in each group of ischemia - reperfusion 9 hours was similar. After ischemia - reperfusion 24 hours , the apoptosis in neural cells in bcl -2 group was reduced significantly.Conclusion1 Injection pLXSN — bcl — 2 after ischemia - reperfusion 3 hours via internal carotid artery may increase expression of bcl -2.2 Injection pLXSN - bcl - 2 after ischemia - reperfusion 3 hours via internal carotid artery may inhibit expression of caspase - 3 and apotosis after ischemia — reperfusion 24 hours.3 Injection pLXSN - bcl - 2 after ischemia - reperfusion 3 hours via internal carotid artery seams that there is no obvious neuroprotection after ischemia -reperfusion 9 hours.
