The Preventive And Therapeutic Effect Of Amiodarone On Ischemia-reperfusion Injury In Rat Hearts In Vivo

IntroductionMyocardial ischemia - reperfusion injury is seriously pathophysiology process which occur after perfusion in AMI. Recently, with the development of all kinds of technology to treat AMI, myocardial ischemia - reperfusion injury has been payed more and more attention. People did a lot of research in basis and clinic to find a effective method which can prevent and treat myocardial ischemia - reperfusion injury. In this study we use amiodarone in ischemic period to interfere in myocardial ischemia - reperfusion injury in rat hearts in vivo. To investigate the preventive and therapeutic effect of amiodarone on myocardial ischemia - reperfusion injury in rat hearts in vivo.Materials and Methods40 wister rats are divided into four groups randomly, contrasting group, ischemia reperfusion group , amiodarone 2. 5mg/kg group and amiodarone 1. 25mg/kg group. The left anterior coronary descending artery was ligated in 10 min, then the ligateing was relieved 30 min. The ECG changes were recorded incessantly from myocardial ischemia to reperfusion and the left ventricle pressure curve were recorded in the end. The ECG was analysed and given score in Lambeth regulation. Myocardial tissue of infracted zone was scissored to measure MDA content and SOD activity.ResultsThe occurrence rate of reperfusion arrhythemias (VF or VT) was 100% in ischemia reperfusion group, score 5. 30 ±0. 82, MDA content was 10. 23 ±2. 23nmol/mgprot, SOD activity was 74. 58 ±31. 09U/mgprot. amiodarone 2. 5mg/kg group have lower rate of reperfusion arrhythemias than the ischemia reperfusion group,score 3. 50 ± 1. 96, MDA content was 6. 83 ±2. 37nmol/mg-prot and SOD activity was 118.43 ±23. 51U/mgprot. The arrhythemia score of amiodarone 1.25mg/kg group was 4. 40 ± 1. 96, MDA content was 8. 15 ±2. 39nmol/mgprot and SOD activity was 87. 72 ±30. 16U/mgprot. There was statistic significance on arrhythemia score , MDA content and SOD activity between the ischemia reperfusion group and amiodarone 2.5mg/kg group.ConclusionAmiodarone has the preventive and therapeutic effect on ischemia — reperfusion injury in rat hearts in vivo.
