IntroductionAlzheimer' s disease is a kind of degeneration disease of nervous system to old people. Its special pathological feature is Aβ deposition outside the nervous cells and amyloid plaque formation, tau protein over phosphorated and neural fibers twist. In recent years , scientists paid more attention to the function of pathological changes of synapse. They found that the loss of synapse is the basis of dementia of AD. Growth - associated protein 43 ( GAP -43) and synapsin are the component of anterior membrance and vesicle of synapse, can reflect the plasticity of nervous system to injury by testing them. We regard them as the trademark of synapse. We will observe the influence of Aβ1(1-40) on synapsin and GAP-43 of PC 12 cells by immunohistochemical methods. We will discuss the possible mechanism of the loss of synapse of AD patients, and provide the experimental data for the prevention and treatment of AD.Materials and methodsWe cultured the PC 12 cells for days and used them for the test when they had good process. They were randomly divided into two groups. Test group:add Aβ1-40;control group: add the same amount of normal saline,other drugs were the same. We set 5 time - points:Oh,2h,4h,6h,12h,and set 6 holes for each time - point. At different time - point, the cells were made immunohistochemical staining. The optical data were measured by micro - image analysis system. We used statistical software for analysis at last.Result1. The express of two synaptic protein of test group and control group at different times-.When optical data compared at different time of control group,the difference had no significance. ( P > 0. 05). For the test group, the optical data decreased obviously after 4 hours of giving Apt AQ, and had statistical significance compared with other time - point (P <0. 01). The two synaptic protein almost have no expression after 12 hours of giving A{$, 4Q. At the same time -point, the optical data of test group is less than that of the control group(2 -12h) ,the difference had statistical significance(P <0. 01).2. The tendency of synapsin and GAP -43 of test group at 0 - 12h.The curve of the two synaptic protein is decreased obviously at 2 - 4h, and has statistical significance when compared with control group (P<0.01).ConclusionAfJ can induce the decreased expression of synapsin and GAP —43 of PC 12 cells , which can provide some experimental data for the mechanism of injury of synapse.
