Objective To investigate the therapeutic effect of 125I interstitial brachytherapy seeds on the hepatocellular carcinoma of mice and to analyse the influence of doses of radiation to tumor, its surrouding tissue and the blood cells.Methods Hepatocellular carcinoma were produced with H22 cells on the lateral subcutaneous back in seventy mice, and fifty mice elected were divided into four treatment groups and a control group. In each treatment group, 125I seeds were implanted into the tumor according to the activity, namly A,B,C, D group. Whereas in the control group the dummy seeds were implanted. The survival rates of mice and the change of tumor dimensions were adopted to evaluate the anti-tumor activity of I interstitial brachytherapy seed. Routine pathological slides of tumor tissues were observed under light microscope to evaluate the degree and range of tumor tissues damaged by I seeds. The blood cells of mice were also observed before and after treatment.Results The survival rates of groups A,B,C,D were 80%,80%,60%, 60% respectively, while that of the control group was 40%.The average volumes of tumors in groups A,B,C,D were (203. 8 ± 76. 5)mm3, (235. 7±88. 7)mm3, (518. 2 ±178.4) mm3, (965.5 ± 310.3) mm3 respectively , while that of the control group was (3242.4 ± 879.8) mm3(P <0. 01). Pathological examination showed dramatically coagulative necrosis and degenerative necrosis of tumor cells at the periphery of active 125I seeds. No significant difference of the blood cells in mice were observed before and after treatment(P>0. 05).Conclusion The anti-tumor activity of interstitial brachytherapy with 125I seeds is achieved in hepatocellular carcinoma of mice with little side effects. More importantly,the therapeutic effects is well dependent on the peripheral dose of the tumors.
