Objective: At present, the new trend of the tumour's treatment is biological therapy. As the antigen of tumours is hidden, the effect of cytotoxic activity of the effector cells is not strong. To expose the hidden antigen is an important aspect of the biological therapy of tumors , then the effector cells can find much more killing targets,so we can improve the effect of cytotoxic activity on the tumour. To study the situation of the cytotoxic activity of CIK cells induced by dendric cells pulsed with antigen on the gastric carcinoma cells modified by newcastle diease virus. Methods: Mononuclear cells were isolated from peripheral blood of the healthy adult with ficoll solution. CIK cells and DC cells from PBMC were induced with different cytokines.The proliferation ability of CIK cells was measured by calculation , the phenotype of CIK,DC cells was analyzed by flow cytometer. We will induce DC with thawed gastric carcinoma antigen .The experiment includes four groups.â‘ Group CIK: CIK+SGC -7901.â‘¡ Group DC-CIK: DC-CIK +SGC -7901.â‘¢ Group CIK+NDV: CIK+SGC -7901 +NDV.â‘£ Group DC-CIK+NDV: DC-CIK +SGC -7901 +NDV. At the same time, we wll set up the contrast groups of target cells (SGC -7901) and efecttor cells (CIK, NDV, DC-CIK) .the killing tumor cell's activity of CIK cells was tested with MTT method. The DC pulsed with gastric carcinoma antigen was cocultured with the CIK ,then DC-CIK cells was used to kill gastric carcinoma cells modified by newcastle diease virus. At the same time, we will study the effect of 5-FU,DDP on SGC-7901 cells, the killing tumor cell activity of CIK cells,5-FU,DDP was tested with MTT method. Results: ? Group CIK: Cells in CIK, much higher the efftor -target ratio, much stronger cytotoxic activity against SGC-7901 cells, the cytotoxic activity against SGC-7901 cells(46.4% , E:T 20:1) is higher than that of the low efftor -target ratio(42.9% , E:T 5:1),(P<0.01). (2) Group DC-CIK: Under the same efftor -target ratio, DC-CIK group showed a stronger cytotoxic activity against SGC-7901 cells than CIK group's (P<0.01).
