OBJECTIVE To assess the mutation and expression of the p53 gene in in anaplastic large-cell lymphoma (ALCL).We also correlate the expression of two protein induced by wild-type p53 protein ,p21and MDM2.Based on this study we try to exolore the status and functional of p53 in ALCL and can provide the valuable evidence for clinical therapy ofALCL.METHODS In this study, we assessed ALK+and ALK-ALCL tumors for p53 gene alterations using PCR, single-strand conformation polymorphism(sscp) and directsequencing methods. We immunohistochemically assessed ALCL for p53 expression.The apoptotic rate (AR) was assesses using TUNEL technical.RESULTS Three of 36 (8%) ALCL (1/14 ALK~+ 2/22 ALK~-) had confirmed p53 gene mutations. By contrast, p53 was overexpressed in 36 of 55 (65%) ALCL tumors (16 ALK~+ , 20 ALK~-). p21, a target of p53, was expressed in 15 of 31 (48%) ALCL tumors including seven of 15 (47%) p53-positive tumors. p21expression in a subset of ALCL suggeststhe presence of functional p53 protein.we also found that ~Ser15P53and ~ZSR20P53was expressed in 65%and 50%. Apoptotic rate was significantly higher in p53-positive than p53-negative tumors (mean 2.78 vs 0.91%). CONLUDE l.We conclude that the p53 gene is rarely mutated in ALK+ and ALK~- ALCL tumors. Nevertheless, wild-type p53 gene product is commonly overexpressed in ALCL.High levels of p53 protein expression do not correlate with p53 gene mutations in anaplastic large cell lymphoma 2.From study we can find that P53 has its fully functional in ALCL. 3. From the sudy of significant association between P53, p21 and MDM2 suggest that p53 system can be used as a new target for clinical investigational therapy.
