The incidence rate of ovarian cancer is the 3rd of all the female genital tumor,but its mortality is the 1st.On account of the special anatomic structure of the ovary, most patients are in their late stage of disease when they visit the doctors.Although the cytoreductive surgery and chemotherapy of ovarian cancer have being improved,but 5 years survival rate was not increased significantly and it is still the malignant gynecological tumor whose mortality is the highest.So the need of new therapeutical method and drugs is urgent. Interleukin 8(IL-8) is a kind of angiogenesis factor which has great relationship with neogenesis and growth of tumor vessels, progression and metastasis of tumor.The research of targeting IL-8 possess great therapeutic and significance. NF-κB(Nuclear transcription factor-κappaB)is a transcription factor which can upregulate genes expression on progression and metastasis of cancer cells, so NF-κB takes part in the progression and metastasis of carcinoma. Our experiment explored the new way to the therapy of ovarian cancer. We use PDTC to suppress the activation of NF-κB, thereby reduce the expression of IL-8 in SKOV3 cell line.Object:To investigate the change of expression of IL-8 of SKOV3 in acid environment, the role of NF-κB in the process and the effect of PDTC to the proliferation SKOV3 in acid environment and expression of IL-8,which may be the theory basis of using PDTC. Method:To separate SKOV3 cell line to normal culture group,acid environment group,acid environment plus PDTC 25μmol/L,50μmol/L,100μmol/L.We determined the effect of PDTC to the proliferation of SKOV3 in the acid environment by MTT,the content of IL-8 protein in the culture supernatant by ELISA and the expression of NF-κB protein by Western blot. Result: 100μmol/L PDTC could suppress the proliferation of SKOV3 in the acid environment,the rate was as high as 31.65%.The secretion of IL-8 of SKOV3 was increased in the acid environment.After using PDTC,the secretion of IL-8 reduced and the change show dose-effect relation.As the increase of dose,the suppression of PDTC to IL-8 of SKOV3 increased.We observed significant expression of NF-kB in the SKOV3 cells in the acid environment and reduced expression of it after PDTC.Conclusion:Many factors could lead to the activation of...
