Glioma, the most common primary brain cancer, accounts for approximately 50% of all human intracranial tumors. At present, the treatment strategy of glioma is still a cosmopolitan difficult problem. Though surgery is preferred, the remnant invading cells will proliferate and cause recurrence besides the neuro-dysfunction consequently. Radiotherapy is usually recommended for gliomas while it's effect is unsatisfied. Traditional chemotherapy is often of little therapeutic value because the drug can hardly pass the blood-brain barrier. The prognosis of patients with malignant glioma is poor, the life expectancy is usually less than 1 year from the time of diagnosis. Therefore, searching and developing an ideal chemotherapy drug for malignant glioma is mandatory.As2O3 as an anti-tumor agent was developed to treat acute promyelocytic leukemia (APL) in clinical. At present, As2O3 was focused on again, researchers tried it on many solid tumors, and testified its therapy effects on lung, liver and colon cancer. While the anti-glioma effect and the ablitiy of passing through the blood-brain barrier were uncertain. To confirm the new viewpoint, experimental study of its anti-glioma effect in vivo and vitro was performed.1 .Anti-tumor effect of As2O3 on C6 glioma cells in vitro. (1) Identification of C6 glioma cellsGlial fibrillary acidic protein(GFAP) is the major constituent of intermediate filaments in astrocytes, which serves as a marker of glial differentiation. The C6 glioma cell cultured in vitro was identified by immunohistochemistry for GFAP, positive cells(with brown color in kytoplasm) proved to be glial cells. (2) As2O3 inhibit the proliferation of C6 glioma cellC6 glioma cells incubated for 4, 12, 24, 48hr with various concentrations of As2O3 ranged from 0.00, 0.625, 1.25, 2.50, 5.0, 10.0μM, followed by assessment of cell viability using the MTT assay.
