Objective:Chemotherapy is the main cure method of advanced stage pulmonary carcinoma. The effect of hormone in the period of tumor therapy is becoming popular topic in these years. MPA is chiefly administrated in the treatment of gynecological tumor. The purpose of the experiment is to investigate the effect of MPA combination chemotherapy in A549 by MPA+ two chemotherapeutics (Carboplatin or Gemzar) and investigate if chemotherapy combination chemotherapy with MPA can promote the effect of synergism in A549 and accelerate the course of apoptosis and provide experimental proof for clinical application. Methods:Experimental cell are divided into blank group (culture), control group (cell) and experiment group (cell + different factors). Experiment groups are divided into MPA group, chemotherapeutics group and combine group to contrast. Cell profiferation was evaluated by MTT assay. Apoptosis is detected by Situ cell death detection kit. Experiment group are divided into MPA group, chemotherapeutics group and combine group we select Gamzar as chemotherapeutics, put cell into different factors, after several hours detect apoptosis rate ,observe morphologic change of apoptosis cell. Result:1. the growth inhibiting of A549 with Gemzar+MPA (5umol/L) are much better than the one of haplo- chemotherapeutics group, it has statistical significance p<0.01.it can not improve the inhibition ratio of tumor cell by Adding dose. And when we achieve the saturated concentration of chemotherapeutics, MPA dose not continue to raise inhibition ratio. Gamzar+MPA can improve apoptosis rate obviously.? It can not improve the inhibition ratio of tumor cell by carboplatin +MPA. It hasn't statistical significance /*>0.05.3. Using high dose MPA(10umol/L) inhibits the growth of A549 but it has not statistical significance in apoptosis rate. Conclusion:The growth inhibiting of A549 with Gemzar+MPA (5umol/L) are much better than that with the one of haplo- chemotherapeutics group. High dose of MPA dose not better effect, chemotherapeutics unite with low dose of MPA can improve the apoptosis rate. It can't improve the inhibition ratio of tumor cell by carboplatin +MPA. Using MPA alone can inhibits the growth of A549 but the apoptosis rate dose not been change. We can infer that MPA perhaps slow down the growth speed of tumor cell, but itself couldn't kill the tumor cell directly.
