Left to right shunt heart diseases with pulmonary hypertension(PH) indicates a severe phase. During the process of PH, activation and high expression of RAAS is one of the accelerating factors. The clinical therapy of PH is not satisfying at the moment. This experiment is to discuss the effect of Valsartan(hypertensin- II receptor resister) on PH,with the purpose to help therapy of PH. We select 48 male Wistar rat, 67week old,weight between 180200g.The rats are randomly separated to 6 groups: 6 week comparing group(group Ⅰ, n =8 ), 6 week shunting group(group Ⅱ, n =8), 6 week treating group(groupⅢ, n =8 ), 11 week comparing group(groupIV, n =8), 11 week shunting group(group Ⅳ, n =8 ), 11 week treating group(groupVI, n =8). Abdominal aorta and inferior vena cava shunting was produced in rats of shunting and treating group, rats in treatment intragastric perfused with valsartan (40mg· kg-1· d-1). Pulmonary artery mean pressure(PAMP)of each rat was measured by using a right cardiac catheterization procedure,the ratio of right ventricular mass to left ventricular plus septal mass (RV/LV+ S) was detected after 6 , 11 weeks. Sample along the midline of right lung sac and boundary, dye with HE and PCNA.Under the optic microscope observe the slice, calculate median and arteriolar relative mid- membrane thickness with CMIAS. Calculate ratio of sarcous artery to realize the sarcous degree of lung vessels. We discover that pulmonary artery pressure of the shunting group increases obviously than that of the comparing group,pulmonary hypertension come into being. The thickness of right ventricle and hyperplasia of pulmonary artery smooth muscle cell increases. Cubage of pulmonary artery endothelioid cell and smooth muscle cell increases, cavity of the vessels become strictured to obturation. Inner elasticity stratum anomaly thin. Cubage and cell viscera of the smooth muscle cell increases. Indexes of the treating group are laigh than the shunting group but higher than the comparing group(P<0.05). Thereout we consider that Valsartan can significantly lower the pulmonary pressure and mitigate the structural changes of the rats with pulmonary hypertension.
