[Objectives] To study the correlation between FAK as well as FAK py397 expression level and invasion/metastasis, differentiation in colon carcinoma.[Methods] Expression of FAK and FAK py397 was examined in specimens of cancer tissue and resection margin of 45patients by immunohistochemical(S-P) method , cancer tissue and resection margin of 9patients by Western-blot. Semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) method was used to detect FAK expression in the colon carcinoma tissue and the resection margin of 30 patients.[Result] The rate of overexpression of FAK and FAK py397 was obviously higher in cancer tissue than in resection margin.A significant correlation was observed between FAK and FAK py397 overexpression and differentiation, depth of tumor invasion stage and lymph node metastases. However, there was no significant association with age, sex, or presence of distant metastasis. The expression of FAK mRNA was obviously higher in cancer tissue than in resection margin. There was significant difference in between them.[conclusion]FAK and FAK py397 were overexpressed in colon carcinoma .FAK expression correlates with differentiation, Increased FAK and FAK py397 expression can accelerate invasion and lymph node metastasis of the colon carcinoma. The expression of FAK mRNA upregulated in colon carcinoma.FAK was involved in carcinogenesis , invasion and metastases.
