Ezetimibe is the first new type of cholesterol lowering agents called the cholesterol absorption inhibitors (CAIs). It provided significant incremental reductions in LDL-C and TG by selectively inhibiting absorption of cholesterol at the brush border of the small intestine and was well tolerated.In this thesis, racemic ezetimibe has been synthesized with overall yield 17.5% from p-hydroxybenzaldehyde as starting material by a series of chemical process, such as, group protection, condensation, cyclo-addition, hydrolysis, catalytic coupling, reduction and deprotection, etc. The effects of reaction condition such as reaction temperature, reaction time and catalyst have been investigated and optimal reaction condition has been achieved. Moreover, two novel derivatives of ezetimibe have been designed and synthesized. One of the derivatives 17 was synthesized. All these productshave been fully characterized by IR, MS, and H NMR.
