Objective: To observe the effect of bone marrow mesenchymal stem cells (BMMSC) on graft-versus-host disease (GVHD) and graft-versus-leukemia (GVL) in acute lymphoblastic leukemic (ALL) mice after allogenic bone marrow transplantation (allo-BMT) and related mechanism.Methods: Male SCID and inbred strain Kunming mice were used as donor mice. 2×10~6 P388 leukemic cells were injected intravenously into female BABL/C recipient mice on day -5 of allo-BMT. Recipients were divided randomly into either allo-BMT group or co-transplantation group. Mice were transplanted according to a standard protocol. In co-transplantation group, each recipient mice was injected intravenously with 6×10~7 bone marrow cells and 3×10~5 BMMSC obtained after three to four weeks of culture from donor mice. Mice from allo-BMT group did receive injection of allogenic bone marrow cells alone. The morphology and quantity of blood cells of recipient mice were observed at nine o'clock every day after allo-BMT; The impact of BMMSC on CD4~+ and CD8~+T cell in vivo was evaluated by flow cytometry at days 3, 7, 10, 20 after allo-BMT; The general manifestation and pathological changing of GVHD were observed after transplantation; The survival time of recipient mice after allo-BMT was recorded;Results: BMMSC could decrease the quantity of CD4~+ T cell and increase CD8~+ T cell in vivo after allo-BMT. BMMSC could alleviate and delay GVHD. BMMSC could increase obviously the survival time after allo-BMT in ALL mice.Conclusion: BMMSC could alleviate GVHD and maintain GVL after allo-BMT in ALL mice.
