Polypeptide A is a derivation of luteinizing hormone-releasinghormone used for the treatment of a large number of disease related withthe regulation of sexual hormones. At the present, the solid phasesynthesis is applied in produce of leuprorelide acetate. But it isheterogeneous reaction and tends to produce many by products, as a resultthe yield of target peptide reduces dramatically with many complicatecompounds. Thus, the purification of target peptide is a key problem tobe solved, in order to commercialize this product measuring up legalstandard.According the molecular weight difference between the objectivepeptide and the impurities in Polypeptide A crude, the product waspurified by gel chromatography. The best separation conditions, such ascolumn length, sample amount and elution flow rate, were selected withexperimental design using orthogonal table. This method has manyadvantages, such as mild reaction condition, simplification and broadseparation range, but it's also has many disadvantages due to someimpurities of which partition coefficient is similar to the objectivepeptide have similar elution volume and hard to separate, therefore, theHPLC with suitable mobile phase was used for further purification. Theresult, analyzed analytical HPLC, indicates that the gel chromatographyis effective in initial gross separation and further purification canbe made by preparative HPLC.The separation and purification of Polypeptide A is a key step ofpreparation therefore the studies on them has great significances bothin social and economy interest.
