Drug screening has been a key step for drug-discovery. Testing the physiological and pharmacological activity of candidate drugs by using suitable models, method and technology, by which we can value candidate drugs, is a core content during drug screening. Thus, establishing a screening model with high sensitivity, specialty, stability and maneuverability is very important.Tumor necrosis factor alpha(TNF α) is a pleiotropic cytokine that can cause a cytotoxic effect on a number of tumors. TNF-α exerts a wide range of biological activities related to cell grow regulation, virus replication, inflammation, immune modulation, infection shock, autoimmune disease rheumatoid arthritis and diabetes, etc. The drugs which can inhibit TNF- α function such as soluble receptor and antibody of TNF- α have been approved in the treatment of human AIDS, rheumatoid arthritis and inflammatory bowel disease.In this paper, we established a model for primary, secondary and confirmatory screening of TNF- α antagonist from Traditional Chinese Medicine(TCM) library, following cDNA cloning and expression of TNF- α and its receptor. Using this model, we got 32 positive samples after primary screening, 10 positive screening after secondary screening and 12 active fragments after separating compounds.
