Verarum nigrum var. ussuriense Naikai belongs to Veratrum genera of Liliaceae family, which is widely distributed in China. The roots and rhizomes of the plant have remarkable physiological activities and the alkaloids from it have hypotensive and antithrombotic activities. The aerial parts of the plant are not used as drugs, but stilbene derivatives isolated from them have antithrombotic, anti-oxidative and anti-tumor activities. For the purpose of utilizing and protecting the plant resource, the extracting methods for total alkaloids from the hypogeal part of the plant, the isolation and identification of constitutents from the aerial parts, and quantitive analytical methods for stilbene deriveatives in the aerial part by HPLC have been described in this thesis.The paper studies three methods for extracting total alkaloids from the roots and rhizomes of the plant, including extraction with ultrasonic in chloroform basified with ammonia, extraction with ultrasonic in ethanol, and extraction with percolation in ethanol. The first method shows highest yield of total alkaloids, which amounts to 1.084% by ultrasonic for 40 minutes.The constituents from the aerial parts of the plant are investigateed. Four pure compounds (Ⅰ~Ⅳ) are obtained by solvent partition and column chromatography over silica gel, and their structures are elucidated as p-sitosterol(Ⅰ), resveratrol(Ⅱ), 2,3',4,5'-tetrahydroxy stilbene(Ⅲ), 2-(3-hydroxy-2,5-dimethoxyphenyl)-7-methoxy-benzofuran-6-ol(Ⅳ) by MS and NMR analysis. Among them, Compound Ⅱ and Ⅲ is isolated from the aerial part of the plant for the first time, and compound Ⅳ is proved to be a new natural compound.HPLC quantitive analytical methods of stilbene derivatives in the aerial parts are established with resveratrol and 2,3',4,5'-tetrahydroxy stilbene as reference compounds. The content of resveratrol and 2,3',4,5'-tetrahydroxy stilbene are 0.1004‰ and 0.1027‰ respectively. This work provides a basis for the further isolation and application of the plant.
