Background and Aim: Carcinoma is a type of multi-gene disease, and a complex process involved multi-factor, multi-stage and multi-gene mutation, which caused by activation of telomerase and cancer gene, devitalization of suppressor gene. Activation of cancer gene is one of key factors of oncogenesis, but devitalization of suppressor gene is more common and important in genesis and progression of cancer. Recently, we paid more and more attention to Runx3 gene, a new suppressor gene. In our study, we examined the expression of Runx3 gene in human gastric carcinoma to explain its relationship with clinical pathological parameters of gastric cancer. Further to discuss whether Runx3 can be a new target of diagnosis and treatment of gastric carcinoma.Methods: RNA and protein were extracted from 40 gastric carcinoma specimens. The Runx3 mRNA and protein were detected by reverse transcription polymerasechain reaction (RT-PCR) and Western blot. The relationships between Runx3 expression and clinicopathological parameters were analyzed.Results: RT-PCR and Western blot indicate expression of Runx3 in gastric carcinoma were lower than that in corresponding normal stomach tissue(P < 0.01). The lower expression of Runx3 protein and mRNA in gastric carcinoma were correlated with TMN stage and histology type(P < 0.01). Expression of Runx3 mRNA was coincident with its protein expression.Conclusion: The expression of Runx3 gene is down-regulated in gastric carcinoma,suggesting a possible involvement of Runx3 gene in carcinogenesis and progressionof gastric cancer, it may be a new target of diagnosis and treatment of gastriccarcinoma...
