Object:Consenescence contacts with human's life and death tightly. Aging is a complex physiological process. It refers many fields. About his reason , scientists have talked about many theories. Free radicals theory say : The Oxygen free radical may cause human aging, because the cells produce many OFR , it can set off DNA damaged, protein damaged ,lipid damaged, and the structure damaged of tissues and cells. These will make membrane's structure and physiological function confusion. So it is a very important springhead of aging.Mitochondrion are in almost very cells. It can respire , change the energy, and do metabolism. It is basic matter to keep cells actively. In resent years , people find : his structure and his function have many changes while the men grow old. The structure of mitochondrion will fall off. These changes can lead body's function declined. This is another theory of aging.Senescence is normal progress , many researches and clinic experiences say : If you use Chinese drug correctly ,they can stayaging well through many ways. How to use Chinese drug, Traditional Chinese medicine 's theory think: to reinforce kidney mainly, supply yang, yin , blood and qi , live blood , constringe, eject damp ,soothe the nerves et al. This theory is a very important in traditional Chinese medicine. Renshenyangrongtang has these effects such as making qi and blood harmoniously, supplying kidney and spleen. "Find the root of the illness, balance yin yang .of body" We can find that it can make valetudinarian stronger .In the experiment of animals, we find it can postpone aging mice's senescence of stomach and intestine, improve SAM's mucous membrane of intestines, hi order to discuss the reason about Renshenyangrongtang's anti-aging , we choice sub-aging mice. By means of blood serum enzymology, histopathology, modal metrology we can observe histology and mitochondrion liver and brain , study how Renshenyangrongtang effect aging mice. Methods:1 The mice were subcutenously injected with D-galactose induced aging mode. Groups with different herb concentrations were created.2 Observing MDA and SOD of blood serum, discussing the theory of his anti-oxidative damage.3.By histopathology and ultrastructure observation , research how the effect of Renshenyangrongtang on tectology of aging mice.(1).Examining the quantitative analysis (stereology) of mitochondrion,studying the effection of mitochondrion structure byRenshenyangrongtang.(2).Measuring the mitochondrial MDA and the membrane fluidity ofmitochondrion, researching how Renshenyangrongtang effect thefluidity of mitochondrial membrane.(3).Investing deletion of mtDNA, studying how Renshenyangrongtangrepair it.Results:1 Setting up the model of aging mice. The study of the microscopicand electron microscope anatomical changes in sub-aging mice showthat his mitochondrion swells, his protuberance disappears, roughendoplasmic reticulum expandes. The change of blood serumaccords with aging.2.The concentration of serum SOD significantly increased and theconcentration of serum MDA decreased in the model group, themembrane fluidity of mitochondrion and the mitochondrion MDAdecreased. The morphometry of mitochondrion shows that the numberof mitochondrion reduced the hole volume of mitochondrion declined,mitochondrion swelled, the protuberance disappeared, the area ofinner membrane minished.3 Renshenyangrongtang significantly lessened the degree of damage of the aging. The damaged structure of mitochondrion was markedly ameliorated under optical and electric microscopies.4 Renshenyangrongtang can markedly heighten the level of serum SOD and reduce the level of serum MDA (P<0.05) .5 Renshenyangrongtang can significantly reduce the consistency of mitochondrion MDA, markedly increase the fluid of mitochondrion membrane.6 Renshenyangrongtang can change the damage of mitochondrion well, especially in high concentration of Chinese drug .7.Examine the 3867bp deletion of mtDNA, not gain the result. Conclusion:The mice were subcutenously injected with D-galactose induced aging mode. The reasons of Renshenyangrongtang improving old people's weakness are as follows: Firstly, it can significantly heighten the level of serum SOD and reduce the level of serum MDA in order to improve ability of resisting oxidation, slower cell's aging. Secondly, it can maintain the number, the volume, the swelling degree of mitochondrion, protect mitochondrion's function and structure. Thirdly, it can decrease the thickness of mitochondrion MDA, amend the protein's structure of mitochondrion membrane, advancemembrane 's order, maintain the membrane fluidity. We summarizethat Renshenyangrongtang has the protective effect on aging bodyfrom many ways.
