Objective: To establish the rat Parkinson disease (PD) model induced with lipopolysaccharide (LPS) and study LPS-induced injury of dopaminergic neurons.Methods: Lipopolysaccharide was stereostatically injected into uni--lateral substantia nigra of rats. The circling behavior was observed by intraperitoneal-injection of apomorphine during the intervals of 1 , 2 and 4 weeks after injection. Tyrosine hydroxylase (TH) immunohisto--chemistry was used to detect midbrain dopaminergic neurons. RT-PCR was used to detect expression of TH mRNA.Results: The circling behavior ipsilateral to LPS injected-side was induced in some rats 2 and 4 weeks following injection , but not in 1 weeks . The marked loss of TH-positive cells in the injured substantia nigra was identified , especially 4 weeks after LPS treatment. The de--creased expression of TH mRNA in the injured substantia nigra was iden-tified , especially 4 weeks after LPS treatment. Conclusions: LPS intranigral injection could injure the dopaminergic neurons in the substantia nigra. The rat PD model induced with LPS was established successfully.
