Objective: To evaluate the distributions of the human leukocyte antigen (HLA) â…  and â…¡ loci that may contribute to the genetic susceptibility to psoriasis in the northern Chinese, Han, and to analyze the haplotype association of the HLA with psoriasis, and to constract the genetic foundation of diagnosis, treatment and prognosis of psoriasis.Methods: The allelic frequencies of HLA â…  and â…¡ was analyzed with the polymerase chain reaction-sequence special primers(PCR-SSP), in nuclear families that were divided into two groups: one was psoriasis-effect(Ps,27 cases) and the other was healthy control(nine cases). The frequency of allele and genotype was calculated with the direct count method and Hardy-Weinberg test. The significant association between HLA alleles in psoriasis and the control group was assessed by the chi-square test with the corrected P-value(Pc). The linkage disequilibrium between HLA alleles was analyzed by linkage disequilibrium calculating method. Results: 10 HLA-A alleles, 15 HLA-B alleles, 14 HLA-Cw alleles, 12 HLA-DPB1 alleles,12 HLA-DQA1 alleles, 10 HLA-DQB1 alleles, 10 HLA-DRB1 alleles were found out. HLA-B*37 (14.81%:0),-Cw*0602(50.00%:27.78%),-DPB1*0501 (44.44%:27.78%) and -DQB1*0501 (18.50%:0) were positively associated with psoriasis, whereas HLA-B*48(1.85%:16.67%) was negatively associated with psoriasis with statistical significance when compared with the controls.HLA-A*01-B*57,A*30-B*13,B*13-Cw*0602,B*15-Cw*0801,B*37-Cw*0602,B*57-Cw*0602A*01-Cw*0602,A*30-Cw*0602,A*68-Cw*0602,Cw*0602-DPBl*0901,Cw*1202-DPBl*0501,B*13-DRBl*0701,B*37-DRBl*1001,B*57-DRBl*0701,DRBl*0701-DQAl*0201,DRBl*1001-DQAl*0105,DRBl*1501-DQAl*0102,DRBl*0401-DPBl*0501,DRBl*0701-DPBl*0901,DQAl*0102-DQBl*0602,DQAl*0105-DQBl*0501,DQAl*0201-DQBl*0202,DQAl*0201-DQBl*0303,DRBl*0701-DQBl*0202,DRBl*0701-DQBl*0303,DRBl*1001-DQBl*0501,DRBl*1201-DQB1*O3O1,DRB1*15O1-DQB1*O6O2 and DPBl*0901-DQAl*0201 wereidentified as high-risk HLA haplotypes for psoriasis.Conclusions:(1) The distributive characteristic of HLA alleles in psoriasis and normal person in northern Chinese was analysed, and there existed significant differences in some gene site between two groups. The gene polymorphism of HLA alleles in population was confirmed.(2) This is the first study which demonstrated not only the different association between HLA alleles and psoriasis, but also some newly found high-risk haplotype in northern Chinese Han psoriasis.
