ObjectiveSystemic lupus erythematosus is an autoimmune disease,the exactly Patho-genesis of systemic lupus erythematosus is not clear, the commnonly opinion is the function and numbers of T cell subsets is abnormality, in the research on CD244 and CD48, people found that regulation of CD8 + T cell proliferation by CD244/CD48 interactions. Based on these above,we detect CD48 and CD244 on the surface of trage cells in patients with LN through flow cytometer in order to explore the association between CD48,CD244 and LN.Material and Method1. material;flow cytometer, PE — conjugated mouse anti — human monoclonal autibody CD3, Percp - conjugated mouse anti — human monoclonal auti-body CD8, FITC - conjugated mouse anti - human monoclonal autibody CD48 and CD244.2. We studied 25 Lupus Nephritis patients who met the 1982 revised criteria of American Rheumatism Association for systemic lupus erythematosus.3. Clinical and laboratory data came from our case records and results from laboratory examination.4. Method;We collected the blood samples, make response of antien and antibody ,then made the sample into fluid consist of separaed cells. Using flowcytometer, recorded the mean fluorescence intensity of CD48 and CD244 on the surface on target cells. All the cell collection was analyzed by Cell Ques Plot software. Then performed statistic analysis for experimental results.ResultsThe percentages of CD4 + cells decreased in LN patient in active group compared with normal control group. The percentages of CD8 + cells significant increased in LN patient in active group and unactivestages compared with normal control group. The ratio of CD4+ to CD8+ T cells(CD4/CD8 ratio) significant decreased in LN patient in active group and unactive group compared with normal control group.The mean fluorescence intensity of CD48 on the surface of CD8 + Tcell in active Lupus Nephritis patients has significant statistic difference compared with normal control group,but there is no significant statistic difference in CD244.The mean fluorescence intensity of CD48 and CD244 on the surface of CD4 + T cell in Lupus Nephritis patients has no significant statistic difference compared with normal control group.The mean fluorescence intensity of CD48 on the surface of CD8 + T cell in active Lupus Nephritis patients is relativity with SLEDAI.CONCLUSION1. The percentages of CD4 + T cells decreased in LN patient in active stages compared with normal control grop.2. The percentages of CD8+ T cells significant increased in LN patient in active stages and unactivestages compared with normal control grop.3. The ratio of CD4+ T to CD8 + T cells( CD4/CD8 ratio) significant decreased in LN patient in active stages and unactive stages compared with normal control grop.4. The mean fluorescence intensity of CD48 on the surface of CD8 + T cell in active Lupus Nephritis patients has significant statistic difference comparedwith normal control grop. the mean fluorescence intensity of CD48 on the surface of CD8 + T cell in active Lupus Nephritis patients is relativity with SLEDAI. We found that there is relationship between CD48 and LN.5. We found that there is no relationship between CD244 and LN.
