Objective: To investigate the mechanism of renal interstitial fibrosis after unilateral uraterla obsteruction and renoprotctive effect of angiotensin Ilreceptor antagonist valsartan on renal interstitial expression of transforming growth factor(TGF)-β1, matrix metalloproteinases(MMP)-9, tissue inhibitors of metallopro - trinases(TIMP)-1.Methods: 36 Wistar rats were randomly assigned to operation group , valsartan treatment after operation group and shame operation group after unilateral uraterla obsteruction. Renal tissues were examined by light microscopy at the 2nd and 4th week after operation. Immunhistochemistry was applied to measured the expression of transforming growth factor(TGF)-β1, matrix metalloproteinases (MMP)-9 ., tissue inhibitors of metalloprotrinases(TIMP)-l. The mRNA of MMP-9 and TIMP-1 in renal of all 3 growps were measured by RT-PCR.Resute: In parallely compared with shame operation group, the expression of TGF- β1 and TIMP-1 , that in the interstitium of operation group and valsartan treatment after operation group remarkly increased(P<0.05). In comparison with operation group and valsartan group could significantly increase the expression of TGF-β1 and TIMP-1. The expression of MMP-9 in operation group and valsartan treatment group were significantly increased than that in shame operation group (p<0.05), and that is higher in valsartan treatment group than operation group(P<0.05 ).And renal interstitial fibrosis in the operation group is severitier than valsartan treatment group by pathology.Conclusion: TGF-β1, TIMP-1 and MMP-9 play an important effection on renal interstitial fibrosis. Renin angiotensin system plays an important effection on formof renal interstitial fibrosis. Valsartan can reduce expression of TGF-P i, TIMP-1 and increase that expression of MMP-9. Valsartan may protect the function of renal.
