Flow Cytometry Of Th1,Th2 Cell In Peripheral Blood In Allergic Rhinitis And Nasal Polyps

ObjectivThis study detected the expression of IL-4 % IFN- Y in peripheral blood mononuclear cells(PBMC) of allergic rhinitis and nasal polyps,To investigate the correlation between the unbalanced relevse of Thl/Th2 cytokins and allergic rhinitis , nasal polyps.MethodsThe PMA stimulated peripheral blood mononuclear cells(PBMC)were obtained from 30 allergic rhinitis , 30 nasal polyps and 30 normal controls.Flow cytometry was adopted to detect the expression percentage of IL-4 and IFN- Y inCD3+/CD8-Tcell.ResultsThe expression percentage of CD3+CD8-IL-4+Tcell in normal controls was 2.76 ±1.25, of CD3+CD8-IFN-γ+Tcell was 18.8±3.87;of CD3+CD8-IL-4+ Tcell in allergic rhinitis group was 7.95 + 1.81, of CD3+CD8-IFN- Y +Tcell was 10.18 + 2.76;of CD3+CD8-IL-4+Tcell in nasal polyps was 6.46+1.97, of CD3+CD8-IFN-γ +Tcell was 55.12 ±5.57, CD3+CD8-IL-4+Tcell and CD3+CD8-IFN-γ+Tcell in nasal polyps group were significantly higher than that in normal controls (p<0.05). While the percentage of CD3+CD8-IFN- Y +Tcell in allergic rhinitis group was singnificantly decreased than of the controls (p<0.05), of CD3+CD8-IL-4+Tcell weas significantly higher than that in normal controls (p<0.05).ConclusionFor recurrent nasal polyps patients, Thl/Th2 cell are all enhanced. They secrete various cytokins .These cytokins in the peripheral blood may play an important role in the recurrent nasal polyps. For allergic rhinitis patients, Th2 cell and its cytokins are enhanced while Thl cell and its cytokins are inhibited.The unbalancement of Th1/Th2 cytokins in the peripheral blood may play an important role in allergic rhinitis.
