AIM To value the effect of 17-β estradiol on apoptosis and MT1-MMP mRNA expression in endothelial cell under high glucose condition . METHODS The proliferation of EVC-304, which is treated by high glucose and/or 17-beta estradiol in different concentration respectively, was measured by MTT assays. Then the apoptosis of ECV-304 was detected by Flow Cytometric(FCM) analysis and DNA Ladder assays. Finally, MT1-MMP mRNA expression is measured by RT-PCR.RESULTS After ECV-304 had been treated by high glucose(30mmol/L) , its proliferation and mRNA expression of MT1-MMP was inhibited significantly. However , the tendency fall back after 17-beta estradiol had been administrated. Furthermore, this effect of 17-beta estradiol reachs peak at its concentration from 0.01nmol/L to 0.1nmol/L . The apoptosis was only detected in cells which had been administrated by high glucose and 17-beta estradiol at low(0.001nmol/L)and high (1.0nmol/L) concentration by DNA Ladder assays.Conclusion High glucose has strong toxicity on vascular endothelial cells. Moderate level of E2 has protective effect on vascular endothelial cells. E2 can enhance MT1-MMP expression of endothelial cells under high glucose.
