ADME evaluation of TCM is the important subjects to TCM's modernization. The information obtained from transport studies on medicines or active compounds of TCM could help us to understand the complex action of compounds and to predict a variety of events related to characteristic of compounds and absorption studies of compound products could be extremely important to design the most reasonable therapeutic dosing regimes. For certain purpose, transport experiments, performed with danshensu and paeoniflorin in Caco-2 cell monolayer model as a permeation tool, had been made to investigate the transport mechanism and interaction between two compounds danshensu and paeoniflorin.The rapid, reliable and simple model was developed and validated. Caco-2 cell monolayer model was often used to determinate intestinal transport characteristics and anticipate permeability limited compound absorption.Caco-2 cell monolayer model was used to research the bi-direction transport of danshensu and paeoniflorin. The effects of time, concentration and P-glycoprotein inhibitor of danshensu and paeoniflorin were studied. The transport of danshensu and paeoniflorin across Caco-2 cell monolayer model maybe was an active transportation mediated by P-glycoprotein transporter.Finally, the Papp of danshensu and paeoniflorin was investigated while two compounds were co-administrated. The effects of paeoniflorin on Papp of danshensu were tested in two directions across Caco-2 cell monolayers. The effects of danshensu on Papp of paeoniflorin were also studied. So the conclusions were that co-administration of these compounds leaded to a decrease transport from apical side to basolateral side, while the transport in the contrary direction were enhanced. When verapamil which is P-glycoprotein transporter co-administrated, the transport from apical side to basolateral side for both of test compounds was accelerated. The transport in the contrary direction were inhibited.
