The lung carcinoma is a common respiratory system malignant tumor,themorbidity of which is high,at the same time its treatment effect is not good.Lung cancer is the leading cause of cancer-related deaths and in many othercountries around the world. It is also one of the common malignant tumor thatserierously affect people's health. But it is not clear for its pathogenesy up tonow. Recently much reseach sign that it is related betweenHER2/neu/c-cerbB-2 and lung canaer.HER 2/neu/C-erB2 gene is normally residing on long arm ofchromosome #17 and produces HER 2 protein ( 185 kDa ) interacting withTyrosine Kinase, ( an enzyme ) and help cells to divide and grow. HER2/neuoverexpression occurs in many kinds of malignant tumors, and was correlatedwith poor prognosis and therapeutic resistance. From recent study, 25 % oflung cancer had this gene amplified. The amplification of the HER 2/neu genecauses HER 2 protein to be overproduced, therfore, cause the lung cancer cellsto divide and increase in size very quickly. HER2 gene amplification andreceptor overexpression have been associated with adverse tumorcharacteristics and poor prognosis. HER 2/neu/C-erB2 does not express innormal tissue,but overexpression in many carcinoma tissue. Her2/neuoverexpression is involved in some antitumor drugs resistance.There are three methods to detect HER-2: fluorescence in situhybridization (FISH), real time reverse transcription (RT)-PCR, andimmunohistochemistry(IHC). IHC is used to detect HER-2 protein. FISH andPCR are used to detect HER-2 gene.In this study,we used IHC method todetect the expression of HER-2 in 30 human lung cancer and 20 nomal lungtissue, and investigate the relationship between their overexpressions and theclinical pathology of lung cancer.The experiment chooses 30 cases lung cancer tissues and 20 cases nomal lungtissues from China-Japan Union Hospital of Jin Lin Universityrandomly.Among 30 cases,there are 7 Squamous Cell Carcinoma(poorlydifferentiated squamous cell carcinoma) ,21 adenocarcinoma(moderatelydifferentiated adenoma 11 cases, poorly differentiated adenoma 10 cases),2small cell carcinoma.The result of the immunohistochemistry indicats as following: HER2were overexpressed in 9 cases of 30 lung cancer cases,and the 9 cases areadenocarcinoma. HER2 were not overexpressed in other 21 lung cancer casesand nomal lung tissues.Adenosquamous cancer was higher than that in theother pathological types of lung cancer.We can draw conclusions from the experiment as following: HER2 wasnot overexpressed in nomal tissue;HER2 was overexpressed in lung cancertissue and expression of HER2 was no corelation with pathologicaldifferentiation;HER2 was expressed in NSCLC tissue remarkably,especiallyin adenocarcinoma;Overexpression of HER2 in lung cancer tissue wasinvolved in some antitumor drugs resistance and poor prognosis.It may be alsoassociated with the origination and development of lung cancer;Overexpression of HER2 in lung cancer tissue indicates that Herceptin may beavailable on the lung cancer with Overexpression of HER2.
