Leukemic cells have some disturbancein the course of proliferation,differentiation and apoptosis . Theetiopathogenisis and pathogenesy of leukemia is always the hot spotof research. In resent years some research indicates that whenleukemia occurs ,there is metabolic abnormality of trace element.Leukemic cells have some abnormality in the course of proliferationand differentiation. While the trace element has effect on theproliferation and differentiation of cells through regulating thesynthesis of ribonucleic acid and histone.Zinc is a trace nutrientindispensable for life and has many kinds of important functions in thebody: 1. zinc can regulate the synthesis of ribonucleic acid andpotentially modulate cellular signal recognition,second messengermetabolism and the function of protein kinase and protein phosphatase.2. Zinc can protect the cells from injury of free radical, which plays animportant role in protecting cellular structure and maintainingmetabolic balance.3. Many metalloenzymes require zinc as a keystructural component or as a cofactor. To accommodatingconcentration of zinc at suitable level,there must exist sophisticatedregulatory systems to maintain zinc homeostasis.Almost all themovement pathways of zinc are mediated by membrane protein,zinctransporters .It is very significant to study the relationship betweenzinc, zinc transporters and leukemia,to investigate mechanism ofaccommodating concentration of zinc, to understand its application atprecaution, diagnosis and therapy.Expression of hZIP1 and ZnT1 mRNAs of the mononuclearcellin bone marrow was analysed by Reverse Transcription-PolymeraseChain Reaction (RT-PCR). Zinc level in supernatant of bone marrowfrom patients with leukemia were measured by flame atomicabsorption spectrometry(FAAS) expression.In this research expression of zinc transporters hZIP1 and ZnT1mRNAs of the mononuclearcell in bone marrow was detected , zinclevel in supernatant of bone marrow from patients with leukemia wasmeasured,and the relationship between which and proportion of blastcell was analysed. Conclusions: 1. The study demonstrated thatexpression of zinc transporters hZIP1 and ZnT1 mRNAs in bonemarrow cells of health adult and patients with leukaemia is analysedby RT-PCR. 2.Expression of zinc transporters hZIP1 mRNA of themononuclearcell in bone marrow was detected,but between incipientgroup, relapsed group, complete remission group and controlgroup,there is not significant difference.Thus, hZIP1 play role inaccommodating zinc balance inside and outside of cells,but it has notsignificant relevance with zinc regulation when leukeamiaoccurs.3.Zinc transporters ZnT1 has significant relevance with zincregulation of the mononuclearcell in bone marrow when leukeamiaoccurs.The raised expression of ZnT1 caused more zinc influx fromthe inside to the outside of cells .So ZnT1 is the significant gene inregulating zinc level. 4. The result in cell strain K562,HL60 iscorresponding with those in clinical collections. 5. The zinc level insupernatant of bone marrow in incipient group and relapsed groupwere remarkably higher than those in control group and completeremission group. It is related with the raised expression of ZnT1andmore zinc influx from the inside to the outside of cells in incipientgroup and relapsed group. 6.
